地球上的生命有机体到底存在多少人类可利用的抗病毒资源?我们应该如何有效地探索这类资源,造福人类社会?为了更好的探索这类资源,我们提出抗病毒免疫进化的“沙漏”模型(Phil. Trans. R. Soc. B,2024),旨在跨物种探索抗病毒天然免疫的多样性。
前期以地球上种类最丰富的动物物种昆虫尾模型,发现昆虫识别病毒核酸激活广谱的抗病毒天然免疫信号通路的分子机制,打破了传统理论的局限,成果包括:①鉴定果蝇识别病毒核酸的模式识别受体(命名为cGLR1&2)及其产生的抗病毒第二信使,包括一种新的环状二核苷酸3'2'cGAMP以及在哺乳动物中已知的2'3'cGAMP,3'2'cGAMP可调节人STING活性并避免被病毒poxin酶降解(Nature,2021);②证明cGAMP诱发STING和NF-κB依赖的广谱抗病毒机制,包括DNA病毒和RNA病毒(Science Signaling,2020);③创新性地把演化生物学研究手段运用于抗病毒天然免疫研究,发现果蝇属昆虫多样的cGLR受体和两种新的环状二核苷酸2'3'cdiGMP和2'3'cdiAMP,前者具有更强的抗病毒功能(Immunity,2023),后者是在研的潜在哺乳动物疫苗佐剂。受邀撰写综述,总结STING通路在10亿年间从细菌到无脊椎动物再到脊椎动物的发生和演化(Current Opinion in Immunology,2022;Faculty Reviews,2021)。以通讯作者(含共同)在Nature、Immunity和mBio发表论文;以第一作者(含共同)在Science Signaling和 Journal of Virology等发表论文。研究成果得到了Nature、Immunity、Trend in Immunology和Signal Transduction and Targeted Therapy等期刊正面的专栏推介。鉴定并命名了多个基因,被权威数据库收入并引用。主持国家自然科学基金面上项目,作为核心骨干参与国家重点研发计划,获得广东省杰出青年基金和首届广东省科协青年培育等人才项目。受邀在全国果蝇会议做报告及在国际昆虫免疫学会议做海报陈述等。入选国家期刊hLife青年编委,Cell Reports等杂志审稿人
